When DNA is found at a crime scene, many tests may need to run to determine who it belongs to. A lot of the time, only a small amount is found, which sucks because that means less tests can be done. Luckily, a thing called polymerase chain reaction was created! This technique is able to quickly make millions of copies of DNA in 3 simple steps: denaturing, annealing, and extending. To begin, the sample of DNA is put in a tube with nucleotides, primers, and Taq polymerase. •Denaturing → separates the 2 strands of DNA from each other at a temperature of 94°C •Annealing → primers bind to end of DNA at 65-40°C •Extending → Taq polymerase binds to primers and new DNA is synthesized at 72°C However, new DNA can’t be made forever, as the materials run out sooner or later. This progression can be seen with the graph on the last picture. There are 3 phases of amplification, as all good things come in 3s. •Exponential → amount of DNA is doubling •Linear → amount of DNA is less than doubling as reactants are being consumed •Plateau → barely any DNA being made as most, if not all, of the reactants are consumed And that’s it! It usually takes about 30 cycles of amplification before the plateau phase is hit.
