Reflecting on this journey fills me with profound gratitude and awe. Receiving an organ donation is nothing short of a miracle. To think that someone selflessly offered one of their kidneys, and in my case, even their bone marrow, is an act of unparalleled generosity and kindness. Moreover, my transplant journey led me to Massachusetts General Hospital (MGH), where I became part of the Tolerance Study, guided by brilliant minds. What does this mean for me? It means that through the unwavering support of my brother Stephen, who courageously donated his kidney and bone marrow, alongside the expertise of my incredible transplant team and the dedication of countless scientists and doctors, I have achieved something remarkable: tolerance. For years, we have heard that artificial intelligence (AI) is going to make physicians obsolete. In medicine, our training focuses on looking for patterns of symptoms that lead us to a diagnosis. For example, a swollen knee, medial pain, and locking or catching is a medial meniscal tear until proven otherwise. Of all the medical fields, radiology seems to be the most amenable to the practical use of AI, and this session gives just one example where a chatbot can determine which patients, after knee injury, need an MRI to rule out an ACL tear after a knee x-ray. Tryp Therapeutics has unveiled its intentions to commence a Phase IIa clinical trial at Massachusetts General Hospital (MGH) in the United States, focusing on psilocybin-assisted psychotherapy as a potential treatment for irritable bowel syndrome (IBS). This recent development follows the validation of Tryp's investigational new drug (IND) application by the US Food and Drug Administration (FDA). The forthcoming study aims to evaluate the impact of this therapy on individuals with treatment-resistant IBS, specifically targeting those afflicted with persistent abdominal pain and distressing gastrointestinal symptoms. The trial aims to recruit participants aged 21 years and above, diagnosed with IBS and concurrently experiencing anxiety, fatigue, and fibromyalgia. Thirteen years ago, in 2010, world-renowned architect and founding HDF Board member Frank Gehry established the Leslie Gehry Prize for Innovation in Science with a generous gift of $1 million. Named for Frank's daughter to celebrate her many talents and gifts – originality, spontaneity, precision, and rigor – all critical attributes in a scientist, this award recognizes scientists who have made extraordinary contributions to the field of Huntington’s disease research. The thirteen recipients of the Leslie Gehry Prize are all trailblazers in their fields, contributing paradigm-changing work to HD research. This year’s recipient, Dr. Vanessa Wheeler, of Harvard Medical School and Massachusetts General Hospital, has focused on somatic instability as the root cause of HD since the mid-2000s. When a gene called huntingtin expands to reach a certain threshold, the person carrying that expansion will go on to develop HD if they live long enough. Over time, the expansion within the huntingtin gene increases in certain areas of the body – notably, the brain. This biological phenomenon of an increase in genetic expansion is called “somatic instability.” Dr. Wheeler's research laid the groundwork for what has become the most prolific topic in HD research this decade, as understanding somatic instability is thought by many leading scientists to be the route to the cure.
