Now that we know a bit more about DNA itself, we can talk about the different kinds of DNA. Most of the time when talking about DNA, the one that comes to mind is nuclear DNA, but there is also mitochondrial DNA. There are several main differences between nuclear DNA and mitochondrial DNA, so we’re going to take a look at those a bit more closely. 1. The location of the DNA: Nuclear DNA is found within the nucleus of the cell – which is more or less the brain of the cell, and mitochondrial DNA is found within the mitochondria of the cells – which is the powerhouse of the cell. More specifically, the mitochondrial DNA is found within the cytoplasm of the mitochondria. 2. The inheritance pattern: Nuclear DNA is a combination of genetic material from all our ancestors (as each generation creates unique combinations between mom and dad and then passes them to their child). Mitochondrial DNA only comes from our mother’s side. This means that mitochondrial DNA is shared between members of a family and will not be a unique combination. Since mitochondrial DNA is less diverse, it is not used as often in fieldwork or is seen to be of less value, but later on, we will see that this is not always true. 3. Number of STRs: The mitochondrial genome (16,569 base pairs) is much smaller than the nuclear genome (3.3 billion base pairs), therefore there will be fewer regions available that may contain STRs. When looking at DNA to identify someone, the most important parts of the sequence to look at are STRs. This is true for both nuclear DNA and mitochondrial DNA, but there are only 3 main STR regions of interest in the mitochondrial genome, as opposed to anywhere from 13 to 20 STR regions for nuclear DNA. While there are several differences between mitochondrial DNA and nuclear DNA, they both hold value, especially when trying to create a genetic profile for an individual. The mitochondria are often referred to as the powerhouses of the cell. Their main function is to generate the energy necessary to power cells. Present in nearly all types of human cells, mitochondria are vital to our survival. They generate the majority of our adenosine triphosphate (ATP), the energy currency of the cell. The ATP consists of three main structures: the nitrogenous base, adenine; the sugar, ribose; and a chain of three phosphate groups bound to ribose. The phosphate tail of ATP is the actual power source that the cell taps. Available energy is contained in the bonds between the phosphates and is released when they are broken, which occurs through the addition of a water molecule (a process called hydrolysis). Usually, only the outer phosphate is removed from ATP to yield energy; when this occurs ATP is converted to adenosine diphosphate (ADP), the form of the nucleotide having only two phosphates. ATP is able to power cellular processes by transferring a phosphate group to another molecule (a process called phosphorylation).
