☑️ *READ ABSTRACT BELOW:* Part 2: Results: We found that healthy postpartum women had a blunted CAR (p<0.001) corresponding to 84% reduction and 80% lower absolute evening cortisol (p<0.001) relative to non-perinatal healthy women. In the postpartum group, there was a trend-level association between lower CAR and higher scores on the WHO Well-Being Index (WHO-5) (p=0.048) and lower Edinburgh Postnatal Depression Scale (EPDS) scores (p=0.04). Conclusion: Our data emphasize the unique hormonal landscape during the postpartum period in terms of blunted CAR and lower absolute evening cortisol in healthy women early postpartum compared to non-perinatal. ☑️ *READ ABSTRACT BELOW:* The sheer number of gene variants and the extent of the observed clinical and molecular heterogeneity recorded in neuropsychiatric disorders (NPDs) could be due to the magnified downstream effects initiated by a smaller group of genomic higher-order alterations in response to endogenous or environmental stress. Chromosomal common fragile sites (CFS) are functionally linked with microRNAs, gene copy number variants (CNVs), sub-microscopic deletions and duplications of DNA, rare single-nucleotide variants (SNVs/SNPs), and small insertions/deletions (indels), as well as chromosomal translocations, gene duplications, altered methylation, microRNA and L1 transposon activity, and 3-D chromosomal topology characteristics. These genomic structural features have been linked with various NPDs in mostly isolated reports and have usually only been viewed as areas harboring potential candidate genes of interest. The suggestion to use a higher level entry point (the 'fragilome' and associated features) activated by a central mechanism ('stress') for studying NPD genetics has the potential to unify the existing vast number of different observations in this field. This approach may explain the continuum of gene findings distributed between affected and unaffected individuals, the clustering of NPD phenotypes and overlapping comorbidities, the extensive clinical and molecular heterogeneity, and the association with certain other medical disorders. Longitudinally, higher scores for hyperactivity at age 5 significantly predicted higher callous-unemotional traits at age 14. Further, objective sleep-EEG measures showed that a higher sleep efficiency and a shorter sleep latency was associated with lower scores for callousness. Cross-sectionally, higher scores for callous-unemotional traits were associated with higher insomnia and stress, while lower insomnia was associated with higher empathy. Mental toughness was unrelated to callous-unemotional traits and empathy. Conclusions: It appears that hyperactivity traits and negative peer relationships and more unfavorable objective sleep patterns at 5 years predicted socially discouraged callous-unemotional traits and low empathy during adolescence. Further, cross-sectionally at the age of 14, callous-unemotional traits, subjective poor sleep, and higher stress were associated. Medieval theories of moral reasoning have their origins in the moral theology of St. Augustine and the rational ethics of Aristotle. For Augustine morality demands the human will’s conformity to the prescriptions of the immutable, necessary and eternal law. ☑️ *READ ABSTRACT BELOW:* Colorectal cancer is the third most common cancer in the world, with an annual incidence of 2 million cases. The success of first-line chemotherapy plays a crucial role in determining the disease outcome. Therefore, there is an increasing demand for precision medicine to predict drug responses and optimize chemotherapy in order to increase patient survival and reduce the related side effects. Patient-derived organoids have become a popular in vitro screening model for drug-response prediction for precision medicine. However, there is no established correlation between oxaliplatin and drug-response prediction. Here, we suggest that organoid culture conditions can increase resistance to oxaliplatin during drug screening, and we developed a modified medium condition to address this issue. Notably, while previous studies have shown that survivin is a mechanism for drug resistance, our study observed consistent survivin expression irrespective of the culture conditions and oxaliplatin treatment. However, clusterin induced apoptosis inhibition and cell survival, demonstrating a significant correlation with drug resistance. This study's findings are expected to contribute to increasing the accuracy of drug-response prediction in patient-derived adenomatous polyposis coli (APC) mutant colorectal cancer organoids, thereby providing reliable precision medicine and improving patient survival rates. Pasquale Guadagno ha 27 anni, sua sorella Annamaria 31, e da quasi 15 anni lottano affinché siano riconosciuti i loro diritti di orfani di femminicidio. Il loro più grande desiderio è che la memoria della madre, Carmela Cerillo, uccisa nel 2010 dal marito Salvatore, venga tutelata. Salvatore Guadagno è stato condannato “con rito abbreviato e l’attenuante della gelosia” a 18 anni di carcere, poi diventati 13 per buona condotta. Il 25 febbraio di quest’anno è uscito dal carcere e ha potuto decidere del corpo della donna che ha ucciso. Carmela avrebbe voluto essere sepolta con la famiglia a Napoli, ma secondo l’assurda legge italiana la decisione di come disporre del corpo spetta al vedovo. Senza l’autorizzazione scritta del padre, i figli non possono fare nulla. Obbligati da questo sistema ingiusto, sono andati in carcere per incontrare l’assassino della loro madre (“l’ultima volta che l’avevo visto mi ha minacciato di morte”, ha raccontato Pasquale), ma lui non ha voluto firmare. Pasquale dice che il padre vuole cremare la moglie, “per tenerla con sé, a casa sua”. E purtroppo i paradossi per gli orfani di un femminicidio non finiscono qui. Dopo l’omicidio, Pasquale e Annamaria non hanno ricevuto alcun supporto, e anzi sono stati affidati alla famiglia paterna, che li ha costretti a visitare in carcere il padre. Una situazione a cui Pasquale si è ribellato a 17 anni andando a vivere a casa della sorella. Non hanno avuto nulla dell’eredità, sia di quello che c’era in casa che nel conto in banca. Quanti orfani sono nella stessa situazione di Pasquale e Annamaria? Magari bambini troppo piccoli per capire cosa sia giusto o sbagliato, e per prendere decisioni. Sono figure di cui lo stato si è completamente dimenticato. E come spesso accade, oltre il danno c’è anche la beffa: i padri omicidi, dopo aver distrutto più di una vita, acquisiscono eredità e diritti sul corpo della vittima. Sowohl die totalitär zu nennenden Maßnahmen während der letzten Fake – „Krise“, als auch die selbstzerstörerische Energie- und Außenpolitik dieser Tage haben nichts mehr mit rationalem Handeln oder Schadensbegrenzung zu tun, sondern pendeln zwischen Dummheit, Inkompetenz, Realitätsverweigerung, stoische Ignoranz, Korruptheit, Arroganz, blindem Aktionismus und transatlantischer Nibelungentreue. Doch ist dieser sog. „Staat“ und seine korrupt - medialen Hilfstruppen fest entschlossen, die ihnen verliehene Macht, Pfründe sowie Deutungshoheit mit allen Mitteln zu verteidigen. Dies geschieht inzwischen kaum noch verbrämt unter offener Verletzung rechtsstaatlicher Prinzipien und richtet sich inzwischen nicht mehr nur gegen aktive Kritiker, sondern gegen jeden, der staatliche Verlautbarungen und Maßnahmen, - und seien sie noch so fragwürdig und grundgesetzwidrig, - hinterfragt. Die totalitäre Anmaßung der Akteure beschränkt sich dabei längst nicht mehr auf den politmedialen Bereich, sondern greift zunehmend in sämtliche Lebensbereiche bis hin zum Privatleben ein. Ebenfalls werden die Methoden einer perversen Umerziehung immer bizarrer, - als stünden die Akteure in einem Wettbewerb, Orwells „Neusprech“ noch zu überbieten. Es überschreitet immer öfter die Grenzen zur Absurdität, wie die ultimative und durch Gerichtsurteile bestätigte Forderung, sich als weiblich gerierende biologische Männer tatsächlich als Frau zu behandeln und damit vehement die Realität zu verleugnen. Ob Personen, die sich für Kaiser Nero oder ein Eichhörnchen halten, ebenfalls die Unterstützung dieser sog. „Justiz“ bei der Durchsetzung ihrer Befindlichkeiten genießen, ist mir nicht bekannt, aber durchaus vorstellbar. Es könnte ein Vorgeschmack darauf sein, was wir im Rahmen aktueller und künftiger Klimarettungs-, Migrations-, Antirassismus- und Diversitätskampagnen zu erwarten haben. Das ehemals weltweit bewunderte Land der Dichter, Denker, Erfinder und Ingenieure ist in meinen Augen eine Großraumpsychiatrie, in der nicht nur besonders verhaltensauffällige, sondern regelrecht gemeingefährliche Patienten die Herrschaft übernommen haben. . . #staat #politik #gender #deutsch

Training in leadership skills needs to be integrated longitudinally throughout a clinician's career. Models of leadership drawn from organisational theories can provide a conceptual framework for cultivating student leadership qualities during teamwork and the evaluation of emergent outcomes. ☑️ *READ ABSTRACT BELOW:* Stress affects brain serotonin (5HT) and dopamine (DA) function, and the effectiveness of 5HT and DA to regulate stress and emotional responses. However, our understanding of the long-term impact of early life adversity (ELA) on primate brain monoaminergic systems during adolescence is scarce and inconsistent. Filling this gap in the literature is critical, given that the emergence of psychopathology during adolescence has been related to deficits in these systems. Here, we use a translational nonhuman primate (NHP) model of ELA (infant maltreatment by the mother) to examine the long-term impact of ELA on adolescent 5HT1A, 5HT2A and D2 receptor systems. These receptor systems were chosen based on their involvement in stress/emotional control, as well as reward and reinforcement. Rates of maternal abuse, rejection, and infant's vocalizations were obtained during the first three postnatal months, and hair cortisol concentrations obtained at 6 months postnatal were examined as early predictors of binding potential (BP) values obtained during adolescence using positron emission tomography (PET) imaging. Maltreated animals demonstrated significantly lower 5HT1A receptor BP in prefrontal cortical areas as well as the amygdala and hippocampus, and lower 5HT2A receptor BP in striatal and prefrontal cortical areas. Maltreated animals also demonstrated significantly lower D2 BP in the amygdala. None of the behavioral and neuroendocrine measurements obtained early in life predicted any changes in BP data. Our findings suggest that early caregiving experiences regulate the development of brain 5HT and DA systems in primates, resulting in long-term effects evident during adolescence. ☑️ *READ ABSTRACT BELOW:* During intrauterine life, external stimuli including maternal nutrition, lifestyle, socioeconomic conditions, anxiety, stress, and air pollution can significantly impact fetal development. The human brain structures begin to form in the early weeks of gestation and continue to grow and mature throughout pregnancy. This review aims to assess, based on the latest research, the impact of environmental factors on fetal and neonatal brain development, showing that oxidative stress and inflammation are implied as a common factor for most of the stressors. Environmental insults can induce a maternal inflammatory state and modify nutrient supply to the fetus, possibly through epigenetic mechanisms, leading to significant consequences for brain morphogenesis and neurological outcomes. These risk factors are often synergic and mutually reinforcing. Fetal growth restriction and preterm birth represent paradigms of intrauterine reduced nutrient supply and inflammation, respectively. These mechanisms can lead to an increase in free radicals and, consequently, oxidative stress, with well-known adverse effects on the offspring's neurodevelopment. Therefore, a healthy intrauterine environment is a critical factor in supporting normal fetal brain development. Hence, healthcare professionals and clinicians should implement effective interventions to prevent and reduce modifiable risk factors associated with an increased inflammatory state and decreased nutrient supply during pregnancy.
