Joubert Syndrome a rare genetic disorder characterized by a distinctive brain malformation known as the “molar tooth sign,” seen on brain imaging such as MRI. This syndrome affects the development of the cerebellum and brainstem, leading to a range of neurological and developmental features. These may include hypotonia (low muscle tone), breathing abnormalities, ocular motor apraxia (difficulty coordinating eye movements), intellectual disability, and additional physical abnormalities. Joubert Syndrome is caused by mutations in genes involved in the structure and function of cilia, which are hair-like structures involved in cellular signaling and development. This syndrome is typically inherited but can be sporadic. When inherited, there is a wide variety of genes implicated, including CPLANE1, TMEM67, AHI1, and CC2D2A. It typically has an autosomal recessive inheritance pattern. This fusion typically involves the vermis, the central portion of the cerebellum. The condition often coexists with other brain abnormalities, including hydrocephalus, agenesis or hypoplasia of the corpus callosum, and midline facial anomalies. Additionally, it may be associated with developmental disorders like Dandy-Walker malformation. Clinical manifestations of rhombencephalosynapsis vary depending on the severity of associated anomalies but commonly include developmental delay, ataxia, hypotonia, and intellectual disability.
